Here's a report on my first try with PT-141. I bought several from different suppliers - the first I mixed was from Xcel Peptides (
https://www.xcelpeptides.com/product/pt-141/). I bought bacteriostatic water separately and injected 1ml into the 10mg bottle. I noticed the rubber seal was hard to penetrate with the water hypodermic needle; I almost feared breaking the needle.
With no sex on the horizon for the first evening (but hopeful for the next morning), I decided to try just 1mg (i.e. .1ml or 10iu) at 6pm to see what reaction my body had. It was probably a bad time to experiment since I was feeling the effects of a mild cold and some existing lower back pain. But after taking the 1mg of PT-141, my back pain was considerably worse. I'm hesitant to blame the PT-141, especially since it was such a low dose, but it's worth noting. My cold symptoms also became worse, turning into lethargy. I noticed no nausea but also no increase in libido or erections.
There are some reports of positive effects lasting 24+ hours so I was hopeful for lingering positive results the next morning. At 10am, with mid-day sex imminent, I took another 1mg shot (the combination getting to my target dose of 2mg, albeit with a 16 hour delay). I take a daily 10mg Tadalafil and now I added a 50mg Sildenafil. The combination of the two PDE-5 inhibitors is usually, but not always, sufficient to sustain an erection for intercourse so the effectiveness of the PT-141 is impossible to gauge. My main reason for taking the PT-141 is for anorgasmia, not erections. The result was a satisfactory erection, penetration and orgasm. Whether the PT-141 helped is hard to say. Another factor was a 5-day refractory period which usually helps the anorgasmia. Notwithstanding the back pain the night before, the 2mg (spaced 16 hours apart) did not give me any nausea or other intolerable symptoms so I am ready to try a more conventional protocol of 2mg a couple of hours before sex. I may go without the Sildenafil to see how Tadalafil + PT-141 works.
I'd appreciate any comments.
EDIT: Try #2. This time I injected 2mg of PT-141. I quickly felt some slight flushing and slight pressure around my eyes. My normal regimen is, when I have advance notice, to supplement daily 10mg Tadalafil with 50mg Sildenafil an hour before sex. This time I skipped the Sildenafil and relied solely upon the daily Tadalafil and PT-141. Two hours after the PT-141 shot, the fun began. I was feeling no effects from the shot - either positive or negative. My erection was good but I can't say whether that was from the Tadalafil or some extended fun foreplay or the PT-141. It had been a 3-day refractory period which is usually marginal so I was happy for the sustained erection, whatever the cause. Again, in light of the 3-day refractory period, I was pleased with my ability to ejaculate pretty much when I wanted to. (In the past, extended intra-vaginal time would take me past the peak and I would either lose the erection or be unable to cum.) So, once again, I'm not seeing any illl-effects from the PT-141 and while things worked out well, I can't see any direct connection between the PT-141 and the successful outcome. I'm definitely, now 18 hours past the injection, not experiencing raging libido or non-stop erections.
I'm looking for to make sex more enjoyable and reliable. To date, shockwave plus Sildenafil and Tadalafil have helped with the ED. I take Cabergoline for the DE but I don't see much improvement. Any boost in orgasmic response will make it worthwhile. If not, I'll move on to the next thing.
