Freezing TriMix vials

Postby sommers » Tue Apr 21, 2020 2:02 pm

I'm reading that vials can be frozen to prolong life. Is there any danger in the vial exploding (I assume they're glass) like a glass bottle of coke will (expansion of the liquid)
I know I have put a bottle in the freezer to quick cool it only to forget I did it and find out the next day it had exploded in the freezer
thanks for any and all replies!

My vials are not full enough to break from expansion, and they are made of fairly thick glass. They've been frozen a long time with no issues.

When I was using Trimix, it would be shipped 5 vials frozen in a dry ice shipping container with instructions to keep frozen until needed for use.

I used to use Bimix. I would have a monthly vial come in, freeze it, and use it down the road. Out of about 17 vials, only one was bad. By that, when I defrosted the vial, it looked like shattered glass debris was floating inside.

it should not do that. Call the pharmacy and see if they will replace it.
I have an implant now, but when I used trimix, i would draw all of my doses up into syringes (0.4ml) and freeze them. I would pull one out as needed and it would defrost in five minutes or so if i rolled it between my palms.

Any possibility that what you saw was crystallized medication? Some guys here on FT have reported that their stuff crystallized after freezing, and apparently some pharmacists warn of that possibility.

When I used Trimix and Quadmix it was shipped overnight in a Styrofoam box with ice packs. 3 vials would go into the freezer minutes after delivery, one vial left in fridge. I found after 5-6 weeks the one in the fridge would start to lose potency. When I left one Trimix in the freezer for 4 months and defrosted it, the potency was not what it should have been. A tech told me there is one ingredient in Trimix that will lose it's potency within hours if not refrigerated, and refrigeration only slows it down. Freezing slows it down a bit more.
Never had an issue with any vials breaking though.

My thoughts also. However, the site administrator seems to approve.

Only tri needs to be frozen/refrigerated as it is the Alprostadil that degrades.
BiMix travels quite well at room temperature--but Tri/Quad works better for most men...

Bimix
Papavarine
Phentolamine

Trimix
Alprostadil / prostaglandin
Papaverine
Phentolamine

Long-term stability of trimix: a three-drug injection used to treat erectile dysfunction.

Abstract
A three-drug injection mixture of alprostadil 12.5 microgram/mL, papaverine hydrochloride 4.5 mg/mL and phentolamine mesylate 0.125 mg/mL in bacteriostatic 0.9% sodium chloride injection was evaluated for stability at 23 deg C, 4 deg C, -20 deg C and -70 deg C. The injection remained clear and colorless in all samples throughout the study. Chemical stability testing was performed using stability-indicating high-performance liquid chromatographic analysis. Alprostadil was the least stable of the drug components at room temperature and under refrigeration. About 8% alprostadil loss occurred in five days at room temperature; under refrigeration losses of about 6% and 11% occurred after 1 and 2 months, respectively. Frozen at -20 deg C and -70 deg C, less than 5% loss of any of the drug componenets occurred in 6 months. A beyond-use date of 6 months when stored frozen at -20 deg C and 1 month when stored refrigerated at 4 deg C is appropriate for batches of the injection that have passed sterility testing. Room temperature exposure should be limited, and the vial should be returned to refrigeration as soon as possible.
