Consistent dose or increase as vial ages

I have settled in at 15 units. However as the vial approaches 3-4 weeks in the refrigerator I have been increasing to 17.5 units to make up for any deterioration real or perceived. Does anyone else do this ?
63yr old. PDwith 40% dor. Curve. Pre-PD 7”x6”.lost 2 in. due to PD. ED prior to PD. Switched from Sildalifil to Trimix Nov 2023 . Currently titrating Trimix for optimization.

There have been times that I have had needles in the refrigerator for as much a 2 1/2 years, and the results of the injection have been nominally leas than if I prepared the needle the day before. I have frequently had the vial in the freezer for more than a year, and the results have been what I was hoping for. In my 14 years of injecting, I have never thrown any out, and I have had no more than 3 injections that seemed to have poor results due to the age of the medicine. I don't think that I ever added additional units to compensate for the age of the medicine. The only thing that I ever noticed about being in the freezer for extended periods of time was that crystals formed in the vial, but the medicine was still fully effective (I shook it up to try to diminish the crystals).

I get 4 small vials at a time. When first starting I would only use 3-5 units but as it got older I did notice i had to go up a few units to get the same effect. Then once a new vial was used I had to go back down. I do think they lose effectiveness slightly after a time. Currently using 6-7 units up from 3-4 after the vial is about half empty. I keep all mine frozen and only thaw enough to draw into a syringe and then they go right back to the freezer.
50+ yrs old. married 25+ years. hypothyroid, on TRT. 10+ years ED, viagra, cialis now 50% ineffective. now on trimix 2MG phentolamine/30MG papaverine/20MCG alprostadil

I've used six-month-old Trimix kept unfrozen at 40F(4C) without any apparent deterioration. On a vacation I used Trimix kept at room temperature for five days with no diminishment of effect. I've never had to increase the dose to get the same result.

I've never frozen Trimix in seven years of use.
Age 79 in 2024. On testosterone replacement due to hypothalamus malfunction. (Attention depressed guys: low testosterone is a cause.) Healthy health nut but ED due to getting old. Like to keep enough cardiovascular ability to thrust for 30 min.

It is my belief that the more important the alprostadil component is to the effectiveness of your mix the more sensative it will be to non-frozen age. I keep my alprostadil mono-mix frozen and it loses only a very little effectiveness over a year. Unfrozen but refrigerated, alprostadil is supposed to lose effectiveness fairly quickly. And even quicker at room temp.
R.R.P 2011 Mayo Jacksonville, Dr. M. Wehle. Not nerve sparing. C in margins. Radiation 2023, V.E.D, Viagra and PGE-1 (80mcg/ml) injections @ 8 - 14 units. Originally Edex20, then compounded PGE due to cost. Inject. 12 yrs. It works. Treasure coast of FL.

I have found a similar experience...I think it might have to do with the components of Trimix used at the compound pharmacies. I have frozen Trimix and used it after a year with no real drop off in effectiveness at the same dose. And then other times I've noticed a change in it being less effective after like 3-4 months. At first I thought it was my ED getting worse, but then the next batch would stay very effective at 6 months. I just think when using any compound medications there are many variables out of control of the pharmacy and the user. People are people and who knows if a pharmacist left one of the ingredients out of the fridge too long...or their batch is getting close to the expiration date. Some compound pharmacies operate on a thin profit margin..Many things to consider...
70 year old legally separated retired health-care professional. ED began in my early 50's. Viagra worked great for about 5 years, then had cardiac by-pass surgery and eventually moved to injections about 10 years ago.
